Whey stimulates muscle protein synthesis harder and faster than casein. In a controlled trial the gap was about 93 percent at rest and about 122 percent after resistance exercise. That is a large, replicated difference, and it is also a narrower claim than the marketing built on top of it.

Measured over a longer window, casein came out ahead on net protein balance. Here is how both things are true.

The short answer

A 2009 study in the Journal of Applied Physiology gave three groups of young men drinks containing an identical 10 g of essential amino acids as whey hydrolysate, micellar casein, or soy protein isolate, then measured mixed muscle protein synthesis directly using a labelled phenylalanine infusion.

At rest, synthesis rates were 0.091 percent per hour for whey, 0.078 for soy and 0.047 for casein. Whey beat casein by roughly 93 percent. After a bout of unilateral leg resistance exercise the same ranking held and the gap widened: whey was about 122 percent above casein and 31 percent above soy. Soy, despite also being a fast protein, sat between the two.

Why digestion speed is the whole mechanism

Whey and casein both come from milk. They separate because casein clots in stomach acid and leaves slowly, while whey stays soluble and empties quickly. A 1997 study in PNAS tracked what that does to the blood by feeding volunteers milk proteins labelled with carbon-13 leucine.

Whey produced what the authors called a dramatic but short rise in plasma amino acids. Casein produced a prolonged plateau of moderate elevation instead, which they attributed to slow gastric emptying. Nothing about the amino acid content drives that difference. It is a physical property of how the protein behaves once it is swallowed.

The result that reverses the ranking

The same 1997 study measured more than synthesis, and this is the part that rarely survives into a supplement ad. Whey stimulated postprandial protein synthesis by 68 percent against casein's 31 percent, consistent with everything above. But casein inhibited whole body protein breakdown by 34 percent, and whey did not inhibit it at all.

Muscle is a balance of two flows, not one. Because casein slowed the losing side while whey only sped up the gaining side, net leucine balance over the seven hours after the meal was significantly more positive with casein. Same leucine intake in both meals, opposite winner, depending entirely on which window you measure and which side of the balance you count.

Where soy lands, and why it complicates the story

The 2009 trial included a third arm that gets left out of most summaries of it. Soy protein isolate, also a fast protein, produced synthesis rates of 0.078 percent per hour at rest, about 64 percent above casein, and 69 percent above casein after exercise. It sat clearly above the slow protein and clearly below whey.

That middle position matters because it breaks the tidy fast-beats-slow story. If digestion speed alone explained the ranking, two fast proteins should have performed about the same. They did not, and whey still beat soy by 31 percent after resistance exercise. The authors were direct about not knowing which factor was responsible, writing that the differences may come from digestion speed or possibly from small differences in the leucine content of each protein.

Read honestly, that is a study reporting a clean result and an unresolved mechanism in the same paragraph. Anyone selling you a single-sentence explanation of why one powder beats another is more certain than the data.

What that means for someone in maintenance

Acute muscle protein synthesis is a strong short term signal, not a promise about how you will look in six months. It is measured in hours, in a lab, after a single drink, usually in young men. Every step from that measurement to a change in body composition adds noise.

The practical reading is smaller and duller than the percentages suggest. If you are drinking a shake immediately after training, whey's speed is a real, measured advantage on that specific occasion. There is one more reason to hold the percentages loosely. Each of those three groups had six men in it. Eighteen young, healthy participants in total, measured once, in a single session. That is a normal size for a stable isotope study, because the method is expensive and invasive, but it is a fragile base for a rule you intend to follow for years. The direction of the whey and casein finding has held up across repeated studies. The exact size of the gap has not, and should not be treated as a fixed constant.
